The topo world includes various non-radioactive and high-throughput assays for all topoisomerase functions including DNA relaxation, DNA supercoiling, DNA decatenation and ATP hydrolysis.  It also includes DNA cleavage assays for type II topoisomerases including topo II and topo IV in the presence of certain topoisomerase drugs (topoisomerase poisons). The assays are formatted for topoisomerases including human topo I, human topo II, gram-negative and gram-positive bacterial topo I, topo II (gyrase) and topo IV (parC-parE complex).  Furthermore, the principle of the DNA relaxation, supercoiling and decatenation assays are universal and can be used for DNA topoisomerases from any species.  These assays can be used for topoisomerase characterization, drug tests and high-throughput screening (HTS) of topoisomerase inhibitors for drug discovery and research.
